Cytokine secretion in cultured human gingival fibroblasts in response to LPS or gingival overgrowth-inducing drugs

چکیده
Human normal, periodontitis and overgrown gingival fibroblasts were assayed concerning proinflammatory cytokines secreting ability. Stimulating effect of gingival overgrowth-inducing drugs such as phenytoin, nifedipine or cyclosporine A and LPS on the secretion ability of each fibroblast specimen was also estimated. The supernatant of cultured medium was assayed cytokines by ELISA techniques. IL-1B was not detected in any fibroblast specimen. Secretion of IL-6 and IL-8 were similar among the gingival specimens. Secretion of TGF-β was the most dominant among the cytokines in any gingival fibroblast. The largest amount of TGF-Β was observed in overgrown gingival fibroblast. LPS stimulated the secretion of IL-6, IL-8 except of TGF-β. Phenytoin, nifedipine, cyclosporin A stimulated TGF-β secretion about 2 times of baseline in only normal gingival fibroblast. These results suggest that these drugs participate in the gingival overgrowth by secreting TGF-β.

Evaluation of the Role of Herpes Simplex Virus Type 1 in the Secretion of Interleukin-1β (IL-1β) by Cultured Gingival Epithelial Cells
Background and Aims: Several studies have shown that there is greater number of viruses in deep periodontal pockets than in normal gingival sulcus. We also know that virus may have an important role in induction of Il-1β secretion by different human cells. In this study we intended to determine the capacity of Herpes simplex virus type 1 to stimulate Il-1β secretion in human gingival ...
متن کاملOn the Cellular and Molecular Mechanisms of Drug-Induced Gingival Overgrowth
INTRODUCTION Gingival overgrowth has been linked to multiple factors such as adverse drug effects, inflammation, neoplastic processes, and hereditary gingival fibromatosis. Drug-induced gingival overgrowth is a well-established adverse event. In early stages, this gingival enlargement is usually located in the area of the interdental papilla. Histologically, there is an increase in the differen...
متن کاملBacterial antigens induce collagenase and prostaglandin E2 synthesis in human gingival fibroblasts through a primary effect on circulating mononuclear cells.
Our previous work suggests that one mechanism through which connective tissue breakdown might occur in periodontal diseases is the production of metalloproteinases, including collagenase, by gingival fibroblasts. In this study we investigated whether highly purified preparations of lipopolysaccharide (LPS) and lipoteichoic acid (LTA) from a number of putative periodontal pathogens could induce ...
متن کاملPalatal mucosa derived fibroblasts present an adaptive behavior regarding cytokine secretion when grafted onto the gingival margin
BACKGROUND Considering that grafted gingival tissue might have to be adapted to the receptor area and that fibroblasts have the ability to respond to bacterial stimuli through the release of various cytokines, this study investigated whether fibroblasts from the palatal mucosa behave differently when grafted onto the gingival margin regarding cytokine secretion. METHODS Biopsies from the pala...
متن کاملThe Effects of Low Level Laser Therapy on the Expression of Collagen Type I Gene and Proliferation of Human Gingival Fibroblasts (Hgf3-Pi 53): in vitro Study
Background Recent investigations show that both proliferation and secretion of macromolecules by cells can be regulated by low level laser therapy (LLLT). The aim of this study was to determine whether LLLT could induce a bio-stimulatory effects on human gingival fibroblasts (HGF3-PI 53). Therefore, the effect of laser irradiation on human gingival cell proliferation and collagen type I gene ...
